AnovelICE/CED-3proteasewasidentifiedrecently,designatedFLICE2andMch4,andrenamedascaspase-10.Caspase-10hastwodeatheffectordomains(DEDs)thatbindtotheDEDintheadaptermoleculeFADDandrecruitsbothTNFR1andCD95toformcomplexeswiththesereceptors.Caspase-10is,therefore,involvedinCD95-andTNFR1-inducedapoptosis.Caspase-10cleavesandactivatescaspase-3,-4,-6,-7,-8,and-9,therebycausingproteolyticcleavageofmanykeyproteins,suchasPARP.CleavageofPARPoccursinmanydifferentsystemsduringapoptosisandisthehallmarkofprogrammedcelldeath.Caspase-10isexpressedinmanytissuesandcelllines.
