Transglutaminase-3(TG3),anepidermal-typeenzyme,isresponsIBLeforformationoftheepidermis.Duringkeratinocytedifferentiation,TG1andTG3appeartoactcooperativelyinthecross-linkingofproteins,includinginvolucrin,loricrinandsmallproline-richproteins(SPRs).Thesereactionsresultinformationofthecornifiedenvelope(CE),aspecializedcomponentconsistingofcovalentcross-linksofproteinsbeneaththeplasmamembraneofterminallydifferentiatedkeratinocytes.FurThermore,TG3inhairfolliclesisinvolvedincross-linkingstructuralproteinssuchastrichohyalinandkeratinintermediatetohardeningtheinnerrootsheath.Duringdifferentiationintheseprocesses,azymogenformofTG3(77kDa)isactivatedbylimitedproteolysiswithcathepsinsSand/orL.
Thisantibodyrecognizesfull-lengthhumanTG3(77kDa)andtheproteolysedformofhumanTG3(30-47kDa).
