FK506-bindingproteins,FKBP51andFKBP52,arehighmolecularweightimmunophilinsthatarepartofthematureglucocorticoidreceptorheterocomplex.TheN-terminaldomainsoftheseproteinsbindFK506andhavepeptidyl-prolylisomeraseactivitythatconvertsprolylpeptidebondswithintargetproteinsfromcis-totrans-proline.TheC-terminaldomainscontainthetetratricopeptiderepeatsthatareinvolvedinprotein-proteininteractionswithHsp90.
AlthoughFKBP52andFKBP51share~75%sequencesimilarity,theyaffecthormonebindingbyglucocorticoidreceptorsdifferentlyandhavedifferentHsp90-bindingcharacteristics.Inaddition,FKBP51islinkedtotheprogesteronereceptorwhileFKBP52islinkedtoprogesterone,androgen,andglucocorticoidreceptors.
