Heatshockfactor1(HSF1)isaheatshocktranscriptionfactorthatactivatesthetranscriptionofgenesencodingproductsrequiredforproteinfolding,processing,targeting,degradation,andfunction.Up-regulationofexpressionofheatshockproteinsinresponsetostressoccursattheleveloftranscriptionthroughaheatshockelementandaHSFtranscriptionfactor.
AminoacidsequencesformostHSFsarehighlyconserved.ADNAbindingdomainisattheN-terminus,hydrophobicrepeats(essentialtotheformationofactivetrimers)areadjacenttothisbindingdomain,andanothershorthydrophobicrepeat(necessaryforsuppressionoftrimerization)occurstowardtheC-terminus.Inhighereukaryotes,HSF1isdistributeddiffuselyinthecellcytoplasmandnucleusinunstressedcells.Onexposuretoheatshockorotherstresses,HSF1localizestodiscretenucleargranules;onrecoveryfromstress,itreturnstoadiffusenuclear-cytoplasmicdistribution.
Thisantibodyrecognizesan~85kDaprotein(inactiveform)inunstressedcelllysatesandan~95kDaprotein(activeform)inheat-shockedcelllysatesofhuman,mouse,rat,rabbit,bovine,guineapig,hamster,andmonkey.
