Hsp27isanimportantheatshockproteinfoundinnormalandmalignanthumancells.ThebasicstructureofmostHspsisahighlyconservedaminoacidsequencewithanα-crystallindomainattheC-terminusandWD/EPFdomainatthelessconservedN-terminus.TheN-terminusisessentialforformationofhighmolecularweightoligomers.Hsp27oligomersareformedbyasmanyas8-40Hsp27monomers.Thedegreeofoligomerizationisassociatedwithchaperoneactivity:largeoligomershavehighchaperoneactivity,whereasdimershavenochaperoneactivity.Hsp27islocalizedinthecytoplasmofunstressedcellsbutcanredistributetothenucleusinresponsetostresswhereitmayfunctiontostABIlizeDNAand/orthenuclearmembrane.Hsp27isalsoinvolvedintheapoptoticsignalingpathwaybecauseitinterfereswithactivationofcytochromeC/Apaf-1/dATPcomplex,therebyinhibitingactivationofprocaspase-9.
