Hsp90andthe94kDaglucose-regulatedprotein,Grp94,aremajormolecularchaperonesofthecytosolandendoplasmicreticulum.Inmammaliancells,thereareatleasttwoHsp90isoforms,Hsp90αandHsp90β,whichareencodedbyseparategenes.AllknownmembersoftheHsp90familyarehighlyconserved,especiallyintheN-terminalandC-terminalregions.Intheabsenceofstress,Hsp90isanessentialcomponentofcellularprocessessuchashormonesignalingandcellcyclecontrol.

Severalregulatoryproteinssuchassteroidreceptors,cellcyclekinasesandp53havebeenidentifiedassubstratesofHsp90.
