Regulationofα-amino-3-hydroxy-5-methyl-4-isoxazolepropionicacidreceptors(AMPARs)playsakeyroleinalteringexcitatorysynaptictransmissionintheCNS.Interestingly,theregulatorymechanismsdifferbetweendistinctsubunitsofAMPAR,whichrangefromglutamatereceptor1(GluR1)toGluR4(alsoreferredtoasGluA1–4).Forexample,subunitswithalongintracellularcarboxyterminus(i.e.,GluR1,GluR2L,andGluR4)areinvolvedinactivity-dependentsynaptictargetingofAMPAR,whereasthosewithashortercarboxyterminus(i.e.,GluR2,GluR3,andGluR4s)seemtomaintainbasalsynaptictransmission.GluR1hasseveralphosphorylationsitesontheintracellularcarboxyterminus.ManyofthesesiteshavebeendemonstratedtoplayaroleinsynapticAMPARregulationandsynapticplasticity.
