TLR3isa904aaproteinwith24N-terminalleucine-richrepeatmotifs(LRRs)andamolecularweightof97kDa.TLR3ismostcloselyrelatedtoTLR5,TLR7,andTLR8,eachwith26%overallsequenceidentity.Invivo,twodifferentsizedtranscriptsforTLR3areobservedsuggestingthatthemRNAisalternativelysplicedtogeneratetwodifferentformsoftheprotein.TLR3mRNAisexpressedathighestlevelsinplacentaandpancreas.ThereareconflictingreportsregardingtheexpressionofTLR3inparticularleukocytepopulations.SomesuggestthatTLR3isonlyexpressedbydendriticcells(DCs),whileothersfindthatTLR3isexpressedbyTorNKcells.WhileitisgenerallyassumedthatTLRsareexpressedonthecellsurface,itnowappearsthatsome,suchasTLR3,maybelocalizedintracellularly,perhapsinthelysosomalcompartment.
