BACKGROUNDTheTATA-bindingprotein(TBP)playsacentralroleintheassemblyofmosteukaryotictranscriptioninitiationcomplexes.TBPassembleswithotherproteinstoformuniquemultimericcomplexesforeachofthethreedifferentnuclearRNApolymerases.
TBPhasbeenclonedfromavarietyofspeciesandconsistsoftwodistinctdomains.TheC-terminaldomain(~180aminoacids)
ishighlyconservedamongspecies,whereastheN-terminaldomainvariesconsiderablyinlengthandsequenceamongdifferentspecies.TheconservedC-terminaldomaincontainstheDNA-bindingregionaswellasregionsthatinteractwithpositiveand
negativeregulatoryproteins.InhumanTBP,thenon-conservedN-terminaldomainincludesastretchofglutamineresiduesrangingfrom26to42.SeveralresearchgroupshavefoundanassociationbetweenexpansionofthispolyglutaminetractwithrareformsofspinocerebellarataxiaandotherneurodegenerativediseasesincludingHuntington’sdisease.
